Project description:
Antibodies have been the workhouse for present molecular diagnostics due to their high affinity and specificity as a biological recognition element. Aptamers, which are single stranded DNA, have been proven to perform the same tasks with some key advantages. Good specific aptamers are found by screening a larger pool for the right DNA sequences.
This project aims to use a new unique magnetoresistive sensor platform to characterize aptamer - target interactions as function of temperature and buffer. The sensor platform relies on sensitive real-time measurements of surface-bound magnetic beads using magnetoresistive sensors. The measurements can be used to understand and optimize the strength of the aptamer-target bond as function of temperature and buffer. In addition to providing a basic understanding and a tool to compare different aptamers, it may result in improved conditions for aptamer-based binding to a target.
